高效液相色谱法测定酒醅中的乳酸含量(109)

摘要 采用高效液相色谱法测定浓香型白酒酒醅中的乳酸含量。该方法采用浸提、冷冻过滤、减压蒸馏等前处理方式,通过安捷伦ZORBXSB-C18色谱柱(4.6 mm×250 mm,5 μm)分离,以甲醇和磷酸二氢钾溶液(0.02 mol/L,pH2.1)作为流动相,流速0.2 mL/min,检测波长210 nm,柱温30 ℃,进样量10 μL。在100~800 mg/L范围内呈线性相关,相关系数R2为0.99992,重现性RSD为1.11 %~1.35 %,加标回收率为98.88 %~99.52 %。该方法分析结果较好,差异不显著,能够满足定量分析要求。

英文摘要 The determination of lactic acid content in Nongxiang fermented grains was performed by HPLC. The samples was pretreated by extraction, freezing filtration, and vacuum distillation, then separated by Agilent ZORBX SB-C18 column (4.6×250 mm, 5 μm) with carbinol and KH2PO4 (0.02 mol/L, pH=2.1) as the mobile phase; the flow rate was 0.2 mL/min and the detection wave length was 210 nm; the column temperature was at 30 ℃ and the sample feeding amount was 10 μL. A linear correlation could be observed in the range of 100 mg/L to 800 mg/L, the correlation coefficient (R2) was 0.99992, RSD was 1.11 % to 1.35 %, and the average recovery rate was 98.88 % to 99.52 %. As a result, reliable determination results could be obtained and the difference was not significant, which could meet the requirements of quantitative analysis.
