黄孢原毛平革菌预处理对酒糟酶解效果的影响(81)

摘要 为促进白酒糟生物质的酶解糖化,筛选出适宜的预处理条件,以脱除木质素,提高综纤维素(纤维素、半纤维素之和)保留率为目标,研究了黄孢原毛平革菌(Phanerochaete chaysosporium)生长曲线及预处理时间对酶解效果的影响。研究结果发现:在P. chaysosporium接种量为3 %,培养时间为8 d时,酶解液中还原糖、木糖得率达到最大,分别为60.55 %和21.33 %;综纤维素的保留率和木质素的降解率分别为76.85 %和26.44 %;电镜扫描(scanning electron microscope,SEM)及X衍射(X-ray differaction,XRD)结果显示,酒糟经P. chaysosporium处理后酒糟结构发生明显变化,木质纤维网络结构遭到破坏,表面呈现无规则或形状各异的膨松状,沟壑明显,孔隙率增加,比表面积增大,有利于提高水解酶的可及性。

英文摘要 Distillers grains (DG), composed of unhydrolyzed and unfermented polymeric sugar, is the co-product in distilleries. In this study, the effects of Phanerochaete chaysosporium pretreatment on enzymatic saccharification of distillers grains were explored. The results suggested that, as the inoculating quantity of P. chaysosporium was 3 % and the culture time was 8 d, the yield of reducing sugar and xylose reached the maximum of 60.55 % and 21.33 %, and the holocellulose retention rate and lignin degradation rate were 76.85 % and 26.44 % respectively. Besides, SEM and XRD results revealed that there were significant changes in the structure of distillers grains after P. chaysosporium pretreatment including destroyed wood fiber network structure, irregular puffing state in various shapes, and increasing porosity etc., which were helpful for enzymatic saccharification of distillers grains.
