饮用某白酒对大鼠肾上腺髓质细胞DβH、NET表达的影响(17)

摘要 研究摄入某白酒后,不同剂量和不同时长对大鼠肾上腺髓质细胞内多巴胺-β-羟化酶(DβH)、去甲肾上腺素转运蛋白(NET)表达的影响。将健康成年雄性SD大鼠120只,随机分为正常对照组(NCG)及实验组(EG),实验组又分为4周组(4W)、8周组(8W)、12周组(12W)、16周组(16W)、20周组(20W) 5个时间点,其中每个时间点又分为3个剂量组,即低剂量组(0.8 mL/kg·d)、中剂量组(1.6 mL/kg·d)、高剂量组(2.4 mL/kg·d)。实验组大鼠予以54 %vol董香型白酒灌胃,每日2次,正常对照组不予处理。分别于4 W、8 W、12 W、16 W、20 W时取材,采用免疫组化SABC法、蛋白免疫印迹法检测肾上腺髓质中DβH、NET的表达。结果表明,正常组及各实验组肾上腺髓质细胞DβH、NET阳性产物呈棕黄色或棕褐色细颗粒状,分布于肾上腺髓质细胞胞质中,各组髓质细胞阳性产物定位及免疫染色强度未见明显差异。Western-blotting显示第4 W组、第8 W组、第12 W组、第16 W组及第20 W组DβH、NET表达水平与正常对照组相比,差异无统计学意义(P>0.05)。在本实验设定的剂量和时长内用该食用白酒灌胃后对大鼠肾上腺髓质细胞内DβH、NET的表达无明显影响。

英文摘要 To investigate the change of DβH & NET expression in adrenal medulla cell of rats after intragastric administration of 54 %vol Dongxiang Baijiu, 120 normal adult male SD rats were randomly divided into normal control group (NCG) and experimental group (EG). Then EG was divided into five subgroups according to the time as 4 week group (4W), 8 week group (8W), 12 week group (12W), 16 week group (16W) and 20 week group (20W). And each subgroup was divided into low dose group (group L, 0.8 mL/kg·d), middle dose group (group M, 1.6 mL/kg·d) and high dose group (group H, 2.4 mL/kg·d). Intragastric administration of 54 %vol Dongxiang Baijiu was performed in rats in the experimental group twice a day, and the adrenal tissue was taken at the end of 4, 8, 12, 16 and 20 weeks, and the expression level of DβH and NET was tested by immunohistochemical SABC and Western blotting. The results showed that, DβH & NET immunohistochemical positive products existed finely in granular distribution in adrenal medulla cell cytoplasm, compared with the normal control group, there was no significant change of DβH & NET staining intensity and average optical density in each experimental group. Western blotting results showed that there was no statistically significant difference in the expression level of DβH & NET protein between the experimental group and the normal control group (P>0.05). Within the designed time and dosage in our experiment, intragastric administration of 54 %vol Dongxiang Baijiu had no significant effects on the expression level of DβH & NET in adrenal medulla cell of rats.
