复方葛根饮料对小鼠酒精肝损伤的保护作用(131)

摘要 研究复方葛根饮料对酒精所致的急性酒精肝损伤小鼠的保护作用。将SPF级昆明种小鼠,随机分为空白对照组,模型对照组,复方葛根饮料低、中、高3个不同剂量组(5.17mL/kg·BW、10.34 mL/kg·BW、15.51 mL/kg·BW),建立两组小鼠急性肝损伤模型,第1组为小鼠禁食不禁水12 h,一次性灌胃给药(20 mL/ kg·BW),给药30 min后均以50 %vol乙醇(16 mL/kg·BW)灌胃1次,第2组为各剂量组每日经口灌胃相应浓度的受试物(20 mL/kg·BW),试验第31天除空白组外其余各组给予50 %vol乙醇,将小鼠脱臼处死后,第1组模型检测血清谷丙转氨酶(alanine aminotransferase,ALT)、谷草转氨酶(aspartate transaminase,AST)活性、肝组织乙醇脱氢酶(alcohol dehydrogenase,ADH)活性、甘油三酯(triglyceride,TG)、丙二醛(malondialdehyde,MDA)含量;第2组模型检测血清(alanine aminotransferase,ALT)、谷草转氨酶(aspartate transaminase,AST)活性、肝组织甘油三酯(triglyceride,TG)含量、丙二醛(malondialdehyde,MDA)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-px)、超氧化物歧化酶(superoxide dismutase,SOD)活性及肝脏病理组织学检查。结果表明,与模型对照组相比,两组小鼠模型中,各剂量组均能降低血清中ALT和AST活性,降低小鼠肝组织中的ADH活性,提高小鼠肝组织中GSH-px和SOD活性,降低肝组织中TG和MDA含量;各剂量组中肝脏脂肪细胞变性评分在统计学上有显著性差异(P<0.01),肝细胞脂肪变性程度减轻。复方葛根饮料对酒精性肝损伤小鼠具有明显的保护作用。

英文摘要 In this study, the protective effects of compound pueraria drink on mice with acute alcoholic liver injury were investigated. SPF Kunming mice were randomly divided into blank control group, alcoholic injury model group and compound pueraria drink group (low, medium and high dose group respectively as 5.17 mL/kg·BW, 10.34 mL/kg·BW and 15.51 mL/kg·BW).Two models of acute alcoholic liver injury were established as follows: the first group of mice, after 12 hours of fasting but with access to water, disposable gavage administration(20 mL/kg·BW), 30 min later, administered with 50 %vol ethanol (16 mL/kg·BW); the second group of mice were administered with appropriate concentration of subjects for 30 days. On the 31st day, the mice from all groups except the blank control group were administered with 50 %vol alcohol (16 mL/kg·BW), and then the first group of mice were subjected to dislocation death to assay the activities of ALT and AST in serum, the contents of TG and MDA and the activities of ADH in hepatic tissue. The second group of mice were subjected to dislocation death to assay the activities of ALT and AST in serum, the contents of TG and MDA and the activities of GSH-px and SOD in hepatic tissue, and the pathologic examination of hepatic tissue. The results suggested that, compared with the alcoholic injury model group, the activities of ALT and AST in serum ADH in each compound pueraria drink group presented evident decrease, besides, the activities of ADH decreased significantly, and the activities of GSP-px and SOD in liver tissue increased and TG and MDA content in liver tissue decreased (P<0.01). There was statistically significant difference in liver fat cells degeneration score between the two groups. Accordingly, compound pueraria drink had significant protective effects on mice with alcoholic liver injury.
