酸水解法测定保健酒中的总皂苷含量(96)

摘要 为了开发测定保健酒中总皂苷含量更准确的分析方法,将保健酒中皂苷用反相固相萃取法从酒体中分离出来;所得的皂苷提取物用酸水解后,再用溶剂萃取得到苷元;最后用香草醛-硫酸比色法,以人参皂苷Re为标准,测定总苷元含量。结果表明,人参皂苷元(Re)在6.8~91 μg/ mL范围线性关系良好,y=0.011x+0.012(R2=0.999),平均加标回收率为99.15 %,RSD值为1.91 %,精密度、重复性良好。皂苷经水解后消除了糖基对香草醛显色剂的干扰,提取苷元过程采用液液萃取的方式可将保健酒A中的色素完全除去。方法稳定,可作为保健酒中总皂苷的检测及其活性中药成分含量的质量控制的检测方法。

英文摘要 To find a better method for the determination of total saponins content in healthcare liqueur, saponins were separated from the liqueur by SPME, the isolated saponins were acid-hydrolyzed and then extracted by organic solvent, and aglycones were obtained, then aglycones were measured by vanillin-sulfuric acid colorization method and total saponins content was obtained eventually with ginsenonside Re as the reference standard. The results suggested that Re showed a good linear relationship within the range of 6.8 to 91 μg/ ml, the linear equation was y = 0.011x+ 0.012 (R2=0.999), the average recovery was 99.15 ± 1.9 %, and RSD was 1.91 %. The precision and the repeatability of this method was good. Besides, acid-hydrolysis of saponins could eliminate the interference of sugar on vanillin as well as the pigments from other ingredients in healthcare liqueur.
